This experiment is based on "Color Atlas of Practical Flow Cytometry", edited by Shukui Wang and Zhenying Zhou.
Operation method
Preparation of single-cell suspensions for tissue biopsies and endoscopic specimens taken by endoscopy
Materials and Instruments
Tissue Move 1. Immediately after removal, place the material in a penicillin vial containing a small amount of PBS. For more product details, please visit Aladdin Scientific website.
PBS
Scissors Nylon mesh
2. take another small beaker, the mouth of the cup is covered with a 300-mesh nylon mesh, fixed with a string, and moistened with PBS liquid, take fresh tissue specimens and put them on the nylon mesh; because of the small amount of specimens, especially endoscopic sampling should be taken at least 3 or more pieces.
3. Before operation, first moisten the scissors with PBS liquid, and then start to cut the tissue.
4. first cut a few times, depending on the basic absence of tissue blocks, with the original specimen containing penicillin vials in the PBS solution, rinse the cell homogenate and filtered in a small beaker; if there are still visible tissue blocks, and then scissors cut, and then add the appropriate amount of the PBS is rinsed, until the network of no tissue blocks until. This allows as many cells as possible to be collected.
5. Add fixative or cryopreserve the cells and set aside.
