Preservation and recovery experiments of yeast strains
Preservation and recovery experiments of yeast strains
Yeast strains can be stored at -70°C in 15% glycerol (can survive for more than 5 years) or at 4°C on slants of enriched medium supplemented with potato starch (can survive for 1 to 2 years).

Materials and Instruments
Yeast Move 1. Prepare a 30% glycerol solution by adding 1 ml of glycerol solution to each 15 mm x 45 mm, 4 ml capped freezing tube, gently screw on the screw cap and autoclave for 15 min.2. In order to prepare frozen bacterial solution in a freezing tube, add 1 ml of late logarithmic or early stationary culture solution, mix well, place on dry ice for a certain period of time, and then transfer to a -70℃ refrigerator. 3. When resuscitating the strain, a small amount of cells can be scraped from the cryotube and scratched on the plate, do not melt the entire storage solution in the cryotube.
4. Cells can also be prepared as a bacterial suspension by adding 80 μl of DMOS to the frozen strain and stored at -70°C.
