Summary
This experiment is from the website of Basic Medical Experimental Teaching Center of Guiyang College of Traditional Chinese Medicine.
Operation method
Morphological observation experiment of hyaline cartilage
Move
1. Staining: HE
2. Visualization by the naked eye:
The specimen is a cross section of the trachea with blue "C" shaped hyaline cartilage.
3. Low magnification:
Locate the blue-stained hyaline cartilage, and itemize the following structures:
(1) Cartilaginous membrane: located on the surface of hyaline cartilage (note that it surrounds the entire cartilaginous tissue), composed of dense connective tissue, with more fibers in the outer layer than in the inner layer.
(2) Hyaline cartilaginous tissue: the matrix is stained blue, but the coloring varies in shade, with areas near the chondrocytes stained darker. The morphology of chondrocytes is inconsistent, the cells near the cartilage membrane are smaller, oval, single distribution, and arranged parallel to the cartilage membrane. In the deeper part of the cartilage, the cells were larger, round or oval, and distributed in pairs or groups (i.e., homologous cell clusters).
4. High magnification observation:
(1) Cartilaginous capsule: it is the matrix around the chondrocytes, strongly basophilic, with darker staining.
(2) Chondrocytes: when in the living state, chondrocytes fill the whole cartilage trap inside, after fixation and dehydration, the cells shrink. As a result, a translucent void appears between the cells and the cartilage capsule, which is part of the trap.
