Morphological observation experiments of Pneumocystis carinii sp.

Principle

The large trophozoites of Pneumocystis carinii were variable in shape, 2-8 μm long, with only one dark purple nucleus and light blue cytoplasm after staining with Kirschner's solution. The small trophozoites were 1~1.5μm in size, spherical or amoeboid. Pre-encapsulation, 3~5μm long, egg-shaped, with several filamentous pseudopods, a bunch of mitochondria and 1~8 nuclei were visible in the encapsulation with phase contrast microscopy with 8 ascospores, and the nuclei were visible after special staining.

Encystment: Biopsy of encystment taken from respiratory and lung tissues, spherical, 4-6 μm, with 8 ascospores under phase contrast microscope.
