Experiments on the preparation of suppositories

Summary

Suppositories can be categorized into two types according to their action. One is localized in the lumen; the other is absorbed from the lumen into the bloodstream for systemic action. The preparation of suppositories and the role of play, are closely related to the matrix. Therefore, the selected matrix must meet the quality requirements in order to make qualified suppositories.

Principle

Suppositories can be categorized into two types according to their action. One is localized in the lumen; the other is absorbed from the lumen into the bloodstream for systemic action. The preparation of suppositories and the role of play, are closely related to the matrix. Therefore, the selected matrix must meet the quality requirements in order to make qualified suppositories. I. Preparation of glycerol suppositories
1. PrescriptionGlycerin: 80 gDried carbonate: 2 gStearic acid: 2 gDistilled water: 10 g
2. Method of preparationDried sodium carbonate dissolved in water, add glycerin mixed with a water bath heating, slowly add stearic acid fine powder, with the addition of stirring, to be bubbling boil stops, the solution is clear, poured into the lubricant coated bolus mold (slightly overflowing the mouth of the mold), cold, cut flat, remove the package that is obtained.
3. Notes
(1) For transparent appearance, the saponification must be complete (1-2 hours on the water bath) and the acid should not be stirred too quickly to avoid stirring into the air bubbles.
(2) The amount of alkali is more than 10-15% than the theoretical amount, the saponification is fast, and the finished product is soft and transparent.
(3) The water content should not be too much, otherwise the finished product is cloudy, there are also advocated not to add water.
(4) The bolting mold is preheated to about 80°, cooling is slower, and the hardness of the finished product is more suitable.
II. Preparation of anti-inflammatory pain suppositories

3. Notes
(1) When adding anti-inflammatory pain, if the temperature is too high, it will cause discoloration.

III. Chlorhexidine suppository preparation
1. Prescription

2. Method of preparationTake the prescribed amount of gelatin, place it in a weighed evaporating dish, add 40 ml of distilled water, soak it for about 30 minutes to make it swell and soften, then add glycerin and heat it in a water bath to dissolve the gelatin, continue to heat it so that the weight reaches 36-40 g until it is ready.
Chlorhexidine add polysorbate-80, and mix well, the ice tablets dissolved in ethanol, mixed with the liquid under the stirring and then added to the preparation of glycerin gelatin, stirring evenly while hot into the coated with lubricant vaginal pessary mold (a total of 4), cooled and chipped, removed from the package that is obtained.
3. Notes
(1) Pitai Acetate is mixed with polysorbate-80, otherwise it affects the content of the finished product.
(2) Dissolve ice chips in ethanol.
(3) The finished product shall be a pale yellow transparent vaginal suppository.
(4) Each piece contains 20 mg of chlorhexidine acetate.
(5) In the prescription, polysorbate-80 is a surfactant, which can make chlorhexidine acetate evenly dispersed in the glycerol gelatin matrix.
(6) Glycerin gelatin matrix, with elasticity, and at body temperature does not melt, but slowly dissolved in body fluids to release the drug, so the effect is slow and long-lasting.
