Summary

Autoclaving is the primary means of sterilizing laboratory materials in the laboratory. The main equipment is a steam autoclave, both horizontal and vertical. There are also portable small autoclaves, which are more convenient to use.

The principle of autoclaving is to increase the pressure of steam in the pot and increase the temperature of steam, to achieve the role of killing all microorganisms.

Principle

The principle of autoclaving is to increase the pressure of the steam in the pot and increase the temperature of the steam to achieve the role of killing all microorganisms. Now the most commonly used culture medium sterilization conditions is 1.05kg/cm2 (121 ℃) 15min, too long a time may destroy the nutrients of the culture medium. Sugar-containing biochemical culture medium sterilization conditions is 0.7kg/cm2 (116 ℃) 15 min, or with mobile steam hair sterilization. Sterilization of vessels can be 1.05kg/cm2 (121°C) for 20 min.

Move

The autoclave sterilization operation is as follows:

1. Put the items to be sterilized into the pot, close the door of the pot and tighten the screws. Confirm that it is closed, before entering the next step of the operation.

2. Open the exhaust valve.

3. Turn on the steam switch to deliver steam to the pot or plug in the power supply to generate steam.

4. Observe the exhaust condition of the exhaust valve, and close the exhaust valve when the excluded gas changes from cold air to steam and the temperature is (105℃).

Autoclave sterilization must be noted that the pot must be completely excluded from the cold air before closing the exhaust valve to pressurize, otherwise the sterilization effect can not be achieved.

5. Observe the pressure gauge rises to 1.05 kg/cm2 to start recording time.

6. after the pressure reaches 1.05 kg/cm2, you can adjust the air inlet valve, reduce the amount of air inlet, maintain the pressure and make it stabilize at 1.05 kg/cm2. electric heating. Can cut off the power supply, maintain the pressure for 15min, at most not more than 30min, otherwise the nutrients are damaged.

7. Turn off the air inlet valve or cut off the power supply, so that the contents of the pot to cool down normally, not immediately open the exhaust valve.

8. Wait until the pressure inside the pot drops to zero before opening the door and removing the items.
