Overview

This product lyses anucleated red blood cells based on the principle that the difference in salt ion concentration inside and outside cells causes the cell membrane to swell and rupture. Its main components are EDTA, NaHCO₃ and NH₄Cl. The product is at 1× concentration and can be used directly.

Instructions for Use (For Reference Only):

1. Add 3 volumes of 1× Red Blood Cell Lysis Buffer to 1 volume of fresh whole blood. For example, add 3 mL of lysis buffer to 1 mL of fresh whole blood, then mix gently by vortexing or inverting the tube.

2. Incubate on ice for 15 minutes, and gently vortex to mix twice during the incubation. The solution shall become clear and transparent after complete red blood cell lysis.

3. Cell collection: Centrifuge the sample at 450×g for 10 minutes at 4 °C to pellet leukocytes, then carefully aspirate and discard the supernatant.

4. Add 2 volumes of Red Blood Cell Lysis Buffer to the leukocyte pellet, and gently vortex to fully resuspend the leukocytes. For an initial blood volume of 1 mL, add 2 mL of the lysis buffer.

5. Centrifuge again at 450×g for 10 minutes at 4 °C to pellet leukocytes, then carefully aspirate and discard the supernatant.

6. Resuspend the cell pellet for subsequent experiments: If RNA extraction is required, it is recommended to use solutions prepared with DEPC-treated water starting from this step. (Tissue cell treatment: Digest fresh tissues with trypsin, collagenase or other digestive enzymes to prepare a single-cell suspension. Centrifuge and discard the supernatant, then follow the same procedures described above.)

Precautions:

1. For personal safety and health, please wear a lab coat and disposable gloves during operation.

2. This product is only applicable for lysis of anucleated red blood cells of mammals; it is not suitable for nucleated red blood cells (e.g., avian red blood cells).
