Product description
Goldview is a nucleic acid dye with a green band, but it may be mutagenic to cells like EB. When DNA is detected by agarose electrophoresis, Goldview can produce a strong fluorescent signal after binding to nucleic acid. Its sensitivity is equivalent to EB. The method is exactly the same. Double-stranded DNA exhibits green fluorescence under ultraviolet light, while single-stranded DNA exhibits red fluorescence.
Product parameters: Ex (nm): 492, Em (nm): 526
Instructions
1. Melt 100mL agarose gel solution (concentration is generally 0.8%~2%) in a microwave oven.
2. Add 5μL Goldview and shake gently to avoid air bubbles.
3. Pour the gel when it is not hot, and load the sample for electrophoresis after the agarose gel has completely solidified.
4. After electrophoresis, observe under ultraviolet light or visible light. If you use a gel imaging system to take pictures, by adjusting the aperture, exposure time, and selecting the appropriate filter, you can get a clear image with a low background. If you use a digital camera to take photos, turn off the camera's flash and put it in automatic mode.
Precautions
1. Goldview is a nucleic acid dye with a green band, but it may be as mutagenic to cells as EB. You should pay attention to the same protection as EB during operation.
2. Repeated melting of the agarose gel added to Goldview may have a certain impact on the sensitivity of nucleic acid detection, but it is not obvious.
3. Goldview can better bind to nucleic acids at pH 3.6~7.0, so it is best to use fresh running buffer during electrophoresis.
4. Since Goldview produces green fluorescence, it is not suitable for taking pictures with general monochromatic film. You can use a gel imaging system to save images.
5. Gels containing Goldview are not suitable for gel recovery tests. For the recovery test, please use SYBR Green I nucleic acid dye.
6. Goldview is particularly suitable for the detection of large fragments of DNA (fragments greater than 1kb, the detection sensitivity is equivalent to EB); when the DNA fragment is less than 1kb, the detection sensitivity is lower than EB, especially for fragments less than 500bp, Goldview may have weak brightness Or it cannot be detected. If you want to detect small fragments of DNA, please use SYBR Green I nucleic acid dye.
7. The thickness of the glue should not exceed 0.5cm, too thick glue will affect the sensitivity of detection.
8. Since the Goldview solution is relatively acidic, it will irritate the skin and eyes to a certain extent. Wear gloves when operating.
