Principle

Plants in normal growth conditions, from the tip of the leaf or along the edge of the leaf veins termination part of the water holes to discharge water droplets, this phenomenon is called spitting water, which is a kind of root pressure produced by the normal physiological phenomena. External environmental conditions such as temperature, humidity, soil solution water potential, toxic substances, etc., will affect the vital activities of the plant root system, and thus also affect the plant spitting water activity.

I. Materials, instrumentation and reagents

1. Materials: Rice seedlings.

2. Instrumentation: constant temperature incubator, refrigerator, small petri dish, glass cover.

3. Reagents: 25 % (NH4)2SO4, 75 % ethanol.

Experimental Steps

1. Rice seedling culture

Rice seeds were selected, soaked and germinated at a suitable temperature, then sown into petri dishes and cultivated under light for 4-5d, until the seedlings grew to 3-4cm high, and then set aside.

2. Preparation before experiment

8. Five pots of rice seedlings with uniform growth of about 5 cm were labeled with five treatments: moderate temperature (25-30°C), low temperature (ice), concentrated salt (25 % (NH4)2SO4), toxic substance (75 % ethanol) and drought. All five treatments were covered with glass covers.

3. Treatment of seedlings with different environmental factors

(1) Open the glass cover of the moderate temperature (25-30°C) treatment pots, flick off the water droplets on the leaves, (in case of low temperature, add 25-30°C warm water in the pots), and then cover the glass cover.

(2) Open the glass cover of low temperature, concentrated salt and toxic substance treatment pots, remove the water droplets on the leaves, pour away the water in the pots, and use absorbent paper to dry the water on the surface of the root system carefully, and then add ice cubes, 25﹪ (NH4)2SO4, 75﹪ ethanol in the pots respectively, and then cover the pots with a glass cover.

(3) Drought-treated pots should not be filled with water one day before the experiment for drought treatment, and the same glass cover should be put on the pots during the experiment.

4. Observation and analysis of results

After 30-40 minutes of each treatment, observe whether the leaf tips and leaf margins of the seedlings in each pot spit water, record the results, and briefly explain the reasons for the effect of different external conditions on the spitting phenomenon of plants.
