Through the experiment, master the principle and method of observing the stomatal opening and closing condition of plant leaves by the infiltration method.
Operation method
Plant stomatal infiltration assay experiment
Principle
Various liquids have different wetting power on plant leaves, the stronger the wetting power of the liquid, the easier it is to adhere to the surface of the leaf and penetrate into the stomata. Therefore, different liquids can be used to determine the general openness of the stomata.
Materials and Instruments
Plant leaves Move 1. Take the naturally growing leaves outdoors and put a drop of liquid rock vinegar, anhydrous ethanol, benzene and xylene on either side of the midvein of the back of the leaf in turn (it should be noted that different liquids should be dropped on the similar parts, but not in the same place to drop two liquids). The wetting power of the four liquids are: liquid rock vinegar < anhydrous ethanol < benzene < xylene. For more product details, please visit Aladdin Scientific website.

2. Determine the stomatal opening degree according to the infiltration of various liquids, liquid rock vinegar can be infiltrated as large open, liquid rock vinegar can not be infiltrated but anhydrous ethanol can be infiltrated as medium open, anhydrous ethanol can not be infiltrated and benzene can be infiltrated as slightly open, benzene can not be infiltrated and xylene can be infiltrated as nearly closed. The stomata were completely closed if xylene could not penetrate.
