Determination of cytochrome b5

Summary

Cytochrome b5 is a cytochrome deposited in the endoplasmic reticulum membrane of higher animals. It can be purified from rabbit liver microsomes. Because it is membrane bound, it can be solubilized by treatment with proteases or surfactants. The maximum absorption spectra of the reduced and oxidized forms are 555, 526, 423, and 414 mM, respectively, with a molecular weight of 12,500 and a redox potential of +0.02 V. It can be reduced by the same reductase enzymes in the endoplasmic reticulum membrane via NADH. Reduced type b5, together with enzymes known as cyanoreceptor factors, is involved in the fatty acid unsaturation reaction that converts saturated acyl coenzyme A to unsaturated acyl coenzyme A by oxygen.
