Overview

DNase I is a type of endonuclease that can be found in most cells and tissues, preferring to cleave phosphate diester bonds adjacent to pyrimidines, producing polynucleotides with a 5' end phosphate group and a 3' end hydroxyl group. The smallest average digested product is polytetranucleotide.
PROPERTIES
1. Under the presence of Mg²⁺, DNase I can randomly recognize and cleave any site on any strand of DNA; In the presence of Mn²⁺, DNase recognizes two strands of DNA and cleaves them at almost identical sites;
2. DNase I can hydrolyze various forms of DNA, such as single stranded DNA, double stranded DNA, and even chromatin (its cleavage rate is influenced by histones).
STABILITY
The optimal working range is pH 7-8, and the activity of DNase depends on Ca²⁺ and can be activated by divalent metal ions such as Mn²⁺, Zn²⁺, etc. 5mM Ca²⁺ can protect the enzyme from hydrolysis, while 0.1mM Ca²⁺ can reduce the enzyme inactivation rate to half.
APPLICATION
1. Prepare RNA samples without DNA;
2. Removal of possible DNA contamination in RNA samples prior to RT-PCR reaction, including genomic DNA;
3. Removal of DNA templates after in vitro RNA transcription catalyzed by RNA polymerase enzymes such as T7, T3, SP6, etc;
4. Study DNA protein interactions;
5. Notch translation method for labeling probes;
6. Preparation of a random DNA fragment library;
7. Used in TUNEL cell apoptosis detection to prepare positive controls by splicing DNA.
