Tests for the determination of the rate of respiration of plant tissues by Watt's respirator

Summary

Determination of plant respiration rate by Watt's micro respirometer is a classical method of determination, and the basic principles and techniques of this method for determining plant respiration rate are acquired through experiments.

Materials and Instruments

Plant tissue Sprouting rice seed

Watt's micro respirometer with thermostat Tip tweezers Small measuring cylinder 1 ml pipette Electronic balance

Move

1. Weigh three portions each of 0.5 g of plant tissue.

2. 7 reaction bottles were taken, 1 of which was used as a manometer, numbered, and in order, the main chamber of the reaction bottle was filled with the material to be tested (three replicates of each material) and 0.5 ml of distilled water, 0.2 ml of 25 % KOH solution was added to the central cup and a small piece of filter paper, while in the bottle used as a manometer, only 1 ml of distilled water and 0.2 ml of 25 % KOH solution was added.

3. Connect the reaction bottles to the manometer (with the piston on the manometer open), attach the spring and check for leaks.

4. Place the above loaded respirometers on a thermostatic water bath that has been adjusted for temperature (the piston of the detector should be open), the reaction bottle

The reaction bottle should be fully immersed in the water bath.

5, to be kept warm for 10 ~ 15min, so that the temperature inside and outside the reaction bottle is consistent, adjust the right arm of the liquid level to 150mm, note down the left arm of the liquid level height, that is, close the piston on the manometer, and record the start time, every 10min observation, a total of three measurements. Record the measurement data.

6. After the experiment was completed, the reaction bottle was removed, the vaseline inside the bottle was wiped off with cotton and the material was removed, weighed and its volume measured.

The mouth of the reaction bottle was wiped with gasoline and then put into hot soapy water to soak and wash, then rinsed with tap water, rinsed with distilled water, and put into the oven to dry at 60°C for spare.

7. Record the experimental data in the following table

A - reaction bottle manometer reading

B - change in manometer (difference from initial reading)

h-actual change in reaction vial manometer (corrected for thermobarometer reading)

X-oxygen consumption (μl) (calculated by the reaction bottle constant term X=K-h)
