Overview

Enhanced Antifade Mounting Medium is a type of fluorescence decay-resistant mounting medium designed to slow down fluorescence quenching. It effectively suppresses quenching of all common fluorescent dyes and delivers far superior anti-quenching performance compared with standard antifade mounting media; its protective effect only slightly diminishes after 1 to 2 weeks. Calculated based on a usage volume of 50–100 μL per sample, a 5 mL bottle is sufficient for mounting at least 50 samples. This reagent is for research use only and shall not be applied for clinical diagnosis or other purposes.

Materials to Be Prepared Separately:

1. Glass slides

2. Coverslips

3. Fluorescence microscope

Operating Procedures (For Reference Only):

1. Adherent Cell Samples

(1) Aspirate staining solution after staining is completed.

(2) Drop one drop of Enhanced Antifade Mounting Medium onto the glass slide, then place the cell-attached coverslip over it to ensure full contact between cells and mounting medium, and minimize air bubbles.

(3) Observe under a fluorescence microscope, or store the slide at 4 °C protected from light for 1–2 weeks.

2. Tissue Sections

(1) Aspirate staining solution after staining is completed.

(2) Drop one drop of Enhanced Antifade Mounting Medium onto the tissue section, cover with a coverslip to ensure full contact between the section and mounting medium, and minimize air bubbles.

(3) Examine the tissue section under a fluorescence microscope, or store at 4 °C away from light for 1–2 weeks.

3. Other Specimens

Handle other specimens following the protocols for cell samples or tissue sections accordingly.

Precautions:

1. Anti-quenching performance is relative. Samples should ideally be observed immediately after staining; if immediate observation is unavailable, examine them on the same day.

2. Fluorescent substances are susceptible to quenching. All stained samples should be stored at 4 °C protected from light.

3. Although this medium slows fluorescence quenching, samples still require strict light avoidance after mounting.

4. Do not use this mounting medium for slides stained with rhodamine-conjugated antibodies; polyvinyl alcohol-based fluorescent mounting medium is recommended as an alternative.

5. Wear a lab coat and disposable gloves throughout all operations for personal safety and health protection.

6. Use up the reagent promptly once opened to avoid compromised experimental results.
